Monitor changes in cAMP in the primary cilia with high N.A. fluorescence microscopy. A 5HT6 receptor was used for cilia localization. A ratiometric cilia targeted cAMP sensor is available as well. CMV Promoter.


Materials in the Kit:

• Green Cilia-targeted cADDis cAMP Sensor BacMam

• Sodium Butyrate, 500 mM in H2O

The serotonin receptor 5HT6 localizes to the primary sensory cilium.  It is fused to the cADDis cAMP biosensor so that changes in the cAMP in the cilium can be imaged in living cells.  Because the cilium often moves during experiments, a red and green ratiometric sensor is also available.  High numerical aperture objectives, and careful microscopy, are necessary to measure the changes in this delicate organelle, but there is good evidence that changes in cilia cAMP are important to processes such as insulin secretion (Wu et al., 2021).
