U0264G is a fusion of the complete AKAP79 protein to the cADDis biosensor.

Measure cAMP signaling in real time, in the living cells of your choice, with the AKAP79-targeted cADDis cAMP sensor. Increase in fluorescence indicates increase in cAMP presence. cADDis is genetically-encoded and packaged in a BacMam viral vector for easy transduction into a wide variety of cell types, including primary and iPSC-derived cell lines. Simply add cADDis to your cells and measure robust signals the very next day on standard fluorescence microscopes and automated plate readers.

AKAP79

AKAP79 is associated with receptor desensitization and recycling.  In HEK 293 cells it is associated with β-adrenergic receptors, and it is reported to decorate the plasma membrane and recycling vesicles (Wang et al., 2006).  AKAP 79 has been used previously to position a FRET-based cAMP sensor in the cell (Tenner et al., 2020).  In MIN6 cells this targeted sensor responds quite differently to one targeted with the Lyn SH4 motif that localizes to the plasma membrane (Wachten et al., 2009). This indicates that AKAP 79 positions the biosensor in a specialized portion of the plasma membrane.

In neurons, AKAP 79 localizes to postsynaptic densities in dendrites and interacts with calmodulin (Carr et al., 1992; Westphal et al., 1999).   There is evidence that it is involved in the establishment of long term potentiation (Gomez et al., 2002; Woolfrey et al., 2015).

In arterial smooth muscle cells, AKAP79 associates with particular L-type calcium channels, and in the heart it is involved in excitation-contraction coupling (Ercu & Klussmann, 2018).  In the heart, it recognizes a unique pool of cAMP adjacent to the beta adrenergic receptor at the plasma membrane (Surdo et al., 2017)
