Baculovirus pseudotyped with a SARS-CoV-2 Delta Variant (B.1.617.2) spike protein:

Pseudo SARS-CoV-2 is a BSL-1 baculovirus pseudotyped with SARS-CoV-2 spike protein. Pseudovirus enters the host cell through interactions with ACE2 and other host proteins, then delivers a reporter that expresses bright green fluorescence in the host cell nucleus. When viral entry is successfully blocked, the host fluorescence is decreased.

This Pseudo SARS-CoV-2 Delta version is pseudotyped with a SARS-CoV-2 Delta Variant spike protein, also known as B.1.617.2. The mutations in the Delta Variant are T19R, V70F, T95I, G142D, E156-, F157-, R158G, A222V, W258L, K417N, L452R, T478K, D614G, P681R, and D950N. Spike decorates the surface of the pseudovirus, facilitating entry into cells expressing ACE2. BacMam is high titer and more cost effective to use than lentivirus.

Depending on your cell type of interest, additional proteins like ACE2, TMPRSS2, Neuropilin, or Cathepsin-L may need to be expressed to allow for pseudo SARS-CoV-2  entry. Viruses pseudotyped with the original spike protein and D614G spike protein are also available.

• Sodium Butyrate, 500 mM in H₂O
