Baculovirus pseudotyped with a SARS-CoV-2 spike protein with D614G, E484K, N501Y, and K417N mutations:

Pseudo SARS-CoV-2 is a BSL-1 baculovirus pseudotyped with SARS-CoV-2 spike protein. Pseudovirus enters the host cell through interactions with ACE2 and other host proteins, then delivers a reporter that expresses bright green fluorescence in the host cell nucleus. When viral entry is successfully blocked, the host fluorescence is decreased.

This Pseudo SARS-CoV-2 version is pseudotyped with a SARS-CoV-2 spike protein with D614G, E484K, N501Y, and K417N mutations. A SARS-CoV-2 virus variant originally detected in South Africa, and thought to pose increased transmission risk, has these same mutations. Spike decorates the surface of the pseudovirus, facilitating entry into cells expressing ACE2. BacMam is high titer and more cost effective to use than lentivirus.

Depending on your cell type of interest, additional proteins like ACE2, TMPRSS2, Neuropilin, or Cathepsin-L may need to be expressed to allow for pseudo SARS-CoV-2  entry.
