New AKAP-targeted cADDis Sensors

Most cAMP measurements are global measurements of total cellular cAMP. However when biosensors are targeted to different subcellular domains, they often reveal more nuanced views of very localized cAMP signaling. There are many different AKAP Proteins that are localized to different regions of the cell. These AKAP Proteins bind the regulatory subunits of the PKA enzyme, but they often interact with adenyl cyclases, phosphodiesterases, and PKA substrates. This has led to the idea that they define “signalsomes” in the cell. To measure cAMP changes that occur in these singalsomes we have begun to fuse the cADDis cAMP biosensor to AKAP proteins.
